分子間相互作用解析 ITCの最新トレンド

MicroCal製品のアプリケーションマネージャーのVerna Frascaは、Google Scholarを使用して、2018年6月30日までのITCに関わる論文の調査を行いました。その結果を報告します。

ITCとは ~分子間相互作用解析のゴールドスタンダード~

ITCはタンパク質、核酸、脂質などの生体高分子の分子間相互作用解析のゴールドスタンダードと言われています。また、ただ単に結合の強さが求まるだけでなく、ストイキオメトリーや熱力学パラメータの取得も可能なのが特長です。

ITCの歴史 ~世界で初めてのITCはMicroCal~

実はMicroCalが世界で初めてのITCってご存知でしたか?

時をさかのぼること1987年。アメリカ東海岸のノーサンプトンにあったMicroCal LLC(現 Malvern Panalytical)がOMEGA、というシステムから販売を始めました。(驚くべきことに、まだ現役のOMEGAが存在しています!)その後、1992年にMCS ITC、1998年にVP-ITC(こちらはまだまだ販売中!)、2007年には小容量化に成功したiTC200がローンチされました。2004年には世界初の全自動のAutoITCが開発され、2008年に大幅に改良されたAuto-iTC200がリリースされました。そして2015年、ハードもソフトも進化した、PEAQ-ITC、PEAQ-ITC Automatedの製造、販売を開始しました。

世界各国で何千台というITCがライフサイエンス分野の研究施設と、低分子、およびバイオ医薬品メーカーで使用されています。

ITC論文調査 ~ITCを使用した論文数が飛躍的に増加~

Google Scholarで、過去5年毎に「MicroCal + (ITCの製品名を“or”で羅列)」で検索すると以下のような結果になりました。

MicroCal ITCを使用した論文の数が飛躍的に上昇しているのがお分かりいただけます。

これら論文等を詳しく見てみると、様々なタイプの分子間相互作用を測定しています。例えば、タンパク質―低分子、タンパク質―タンパク質、タンパク質―DNA、抗原―抗体、そして酵素と基質などです。ITCは一般的に、低分子医薬品の候補となる化合物とそのターゲット分子(主にタンパク質)の結合評価に使われています。また、ナノパーティクル、ポリマー、膜、界面活性剤、そしてミセルといった分野の評価も増えてきています。

これらは350を超える出版物に掲載されており、うち、2015年1月から2018年6月末までにMicroCal ITCのシステムを用いた文献が25報以上含まれていたのは以下32の出版物です。(文献数は四捨五入)
