What are lipid nanoparticles?

[LNP-diagram.jpg] LNP-diagram.jpg Lipid nanoparticles are small spherical vesicles formed of a mixture of ionizable cationic lipids, helper lipids (such as DSPC and cholesterol), and PEGylated lipids.

These tiny structures offer immense benefits as a drug delivery system, including:

  • Enabling delivery of a wide range of therapeutic payloads, from small molecules to oligonucleotides
  • Superior safety (relative to viral vectors)
  • Streamlined, cell-free manufacturing methods
  • Potential for rapid manufacturing process scale-up

Lipid nanoparticles have a range of medical applications, the most prominent being delivery of DNA and RNA payloads for prophylactic or therapeutic vaccines, in addition to other therapies.

Overcoming lipid nanoparticle formulation development & manufacturing challenges

To better measure these attributes and support confident formulation decisions, developers and manufacturers need a suite of advanced, orthogonal biophysical characterization methods, including:

Lipid nanoparticle formulation development

Formulating lipid nanoparticle products to optimize their stability and therapeutic efficacy is no easy feat since formulation depends not only on the formulation components but also on the formulation process.

Selecting the right formulation strategy demands deep and accurate insights into the attributes of your lipid nanoparticle samples, including:

  • Vector size
  • Concentration
  • Surface charge
  • Composition
  • Thermal stability

Lipid nanoparticle manufacturing and quality control

Manufacturing consistently high-quality lipid nanoparticle products poses numerous challenges. LNP developers must understand the impact of (and be able to optimize) manufacturing process parameters and be able to rapidly conduct batch-to-batch consistency and stability studies.

What’s more, personalized medicines (which are the focus of many LNP therapies) can make QC testing and analysis much more demanding.

To succeed, LNP developers need deep, rapid, and accurate insights into the attributes of their LNP formulation, including:

  • Size
  • Concentration
  • Composition
  • Thermal stability
